How they differ
Semaglutide is a peptide, and peptides do not survive oral administration without help. Orforglipron is not a peptide at all: it is a small molecule that activates the GLP-1 receptor, designed from the ground up for oral use.
That distinction removes the absorption problem rather than working around it. Where oral semaglutide needs the SNAC enhancer and strict fasting conditions, orforglipron has no such constraints because it is absorbed like a conventional drug.
Handling differs accordingly. A small molecule is far more stable than a lyophilised peptide and is not subject to the same cold-chain and reconstitution requirements.
