How they differ

Tirzepatide binds two incretin receptors, GIP and GLP-1. Retatrutide binds those two plus the glucagon receptor, which is the entire reason it is described as a triple agonist rather than a dual one.

That third arm is the interesting part in the research literature. Glucagon receptor activation is associated with increased energy expenditure, whereas the GIP and GLP-1 arms act mainly on insulin secretion, gastric emptying and central satiety pathways. The three mechanisms are therefore additive rather than redundant.

Tirzepatide has completed the SURPASS and SURMOUNT programmes and is an approved medicine. Retatrutide remains investigational, with no marketed brand anywhere, which is why this catalogue lists a brand equivalent for one and not the other.
