Overview
DSIP (Delta Sleep-Inducing Peptide) is a naturally occurring nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated from the cerebral venous blood of rabbits in 1977 by Schoenenberger and Monnier. Despite its name, DSIP's effects extend beyond sleep modulation — it has been investigated for roles in stress response, neuroendocrine regulation, pain modulation, and free radical scavenging. DSIP is found in both free and bound forms in the hypothalamus, limbic system, and peripheral tissues, and can cross the blood-brain barrier.
Mechanism of Action
DSIP's precise mechanism remains under investigation, but research has demonstrated effects through multiple pathways. It modulates GABAergic and glutamatergic neurotransmission, promotes slow-wave (delta) sleep EEG patterns, and appears to normalise disrupted sleep architecture rather than simply inducing sedation. DSIP also interacts with the HPA stress axis by modulating ACTH and cortisol release patterns. Its antioxidant properties are attributed to upregulation of superoxide dismutase (SOD) and glutathione peroxidase activity.
