Overview
Semax is a synthetic heptapeptide consisting of the ACTH(4-10) fragment (Met-Glu-His-Phe-Pro-Gly-Pro) — the portion of adrenocorticotropic hormone responsible for neurotrophic rather than steroidogenic activity. The C-terminal Pro-Gly-Pro extension protects against aminopeptidase degradation. Developed at the Institute of Molecular Genetics (Moscow), Semax is approved in Russia for cognitive and neurological applications and has accumulated extensive research documentation on neurotrophic factor regulation, neuroprotection, and cognitive enhancement in both animal and clinical models.
Mechanism of Action
Semax stimulates the expression of BDNF, NGF (nerve growth factor), and TrkB receptor in hippocampal and cortical neurons. It activates the MAPK/ERK signalling cascade downstream of tropomyosin kinase receptors, promoting neuronal survival, dendrite branching, and synaptic plasticity. Importantly, Semax lacks the adrenal steroidogenic effects of full-length ACTH — it does not stimulate cortisol production. It also modulates dopaminergic and serotonergic neurotransmission and has demonstrated antioxidant properties through regulation of genes involved in oxidative stress response.
