Overview
GHRP-2 (Growth Hormone Releasing Peptide-2, pralmorelin) is a synthetic hexapeptide GH secretagogue and one of the most potent compounds in the GHRP family. Developed in the 1990s by Bowers and Momany, it was instrumental in the discovery and characterisation of the ghrelin receptor (GHSR-1a) before the endogenous ligand ghrelin was identified. GHRP-2 remains a reference compound in growth hormone research, widely used for its robust and reproducible GH-releasing effects in both preclinical and clinical pharmacology studies.
Mechanism of Action
GHRP-2 activates GHSR-1a on pituitary somatotrophs through a Gq-coupled mechanism involving phospholipase C activation, IP3-mediated calcium release from intracellular stores, and calcium influx through L-type channels. This dual calcium signal triggers GH granule exocytosis. GHRP-2 also acts at the hypothalamic level to stimulate GHRH-releasing neurons and suppress somatostatin release, creating a dual amplification of GH secretion. Unlike ipamorelin, GHRP-2 has notable effects on cortisol, ACTH, and prolactin, reflecting broader activation of hypothalamic pathways including appetite-stimulating circuits.
