Overview
Kisspeptin (metastin) is a family of neuropeptides encoded by the KISS1 gene, with kisspeptin-54 being the full-length form and shorter fragments (kisspeptin-14, -13, -10) retaining biological activity. Discovered initially as a metastasis suppressor, kisspeptin was subsequently identified as the endogenous ligand for the GPR54 receptor (now KISS1R), and its critical role in puberty onset and reproductive function has made it a cornerstone of neuroendocrine research. Loss-of-function mutations in KISS1R cause hypogonadotropic hypogonadism, demonstrating its essential role in reproductive axis activation.
Mechanism of Action
Kisspeptin binds to KISS1R (a Gq/11-coupled receptor) on GnRH neurons in the hypothalamic arcuate and anteroventral periventricular nuclei. Receptor activation triggers phospholipase C signalling, IP3-mediated calcium release, and PKC activation, leading to depolarisation and GnRH pulsatile secretion. GnRH in turn stimulates pituitary gonadotropins (LH and FSH). Kisspeptin neurons integrate metabolic signals (leptin, insulin), photoperiodic cues, and sex steroid feedback, functioning as the central gatekeeper of the HPG axis.
