Overview
SS-31 (D-Arg-Dmt-Lys-Phe-NH2), also known as Elamipretide or Bendavia, is a cell-permeable mitochondria-targeted tetrapeptide developed by Stealth BioTherapeutics. It selectively concentrates in the inner mitochondrial membrane at 1000-5000x its extracellular concentration, independent of mitochondrial membrane potential. SS-31 is one of the most studied compounds in mitochondrial medicine research, with preclinical data spanning cardiac, renal, neurological, and ophthalmic models.
Mechanism of Action
SS-31 selectively binds to cardiolipin, a phospholipid unique to the inner mitochondrial membrane that is essential for optimal function of the electron transport chain. By stabilising cardiolipin-cytochrome c interactions, SS-31 optimises electron transfer, reduces electron leak, and minimises reactive oxygen species (ROS) generation at the source. Unlike conventional antioxidants that scavenge ROS after production, SS-31 prevents excessive ROS formation upstream. This preserves mitochondrial cristae structure and maintains ATP synthesis efficiency.
